Outline of Annual Research Achievements

①前年度同様に、病院内併設可能型BNCT用加速器中性子源装置の実用化のために治療時の線量評価を行う治療計画システム(ツクバプラン)について同集学的療法を実現できるように、ツクバプランをBNCT(中性子)だけでなくX線照射、陽子線照射による線量評価にも対応できるように機能拡張した。また、治療時の線量評価を行う治療計画システム(ツクバプラン)は、モンテカルロコード:PHITSを使用しているが、これの機能を高度化することでデータ保護の安全性を向上させた。これにより、以前の原子炉施設を使った悪性脳腫瘍に対するBNCTでは、BNCTにX線照射を組み合わせた照射を実施していたが、今回の研究成果で正確な占領評価ができるようになり、今後の臨床研究に応用できることが期待される。
②新規ホウ素薬剤などの基礎研究について、味の素株式会社との共同研究で、新規ホウ素化合物の基礎的実験を行った。特に今回用いた新規ホウ素化合物は、腫瘍移行性を高める工夫がなされており、実験結果からも、安全かつ十分な腫瘍滞留 性のある新規化合物を開発することができ、細胞照射、動物照射実験においても良好な治療効果を示すことができた(論文掲載ずみ)。

Reason

①治療計画ソフトの高度化研究開発は順調に進んでいる。
②基礎実験としては2種類の新規化合物について研究が進んでおり、特許化や論文作成につながっている。
③臨床研究について予定よりも若干遅れ気味ではあるが、これはホウ素化合物の供給先からの薬剤がえられていないためであり、入手次第進められると考えてい
る

Strategy for Future Research Activity

①新規ホウ素化合物の開発:新規ホウ素化合物(候補あり、特許申請中)を用いて脳腫瘍細胞や担癌動物、皮下腫瘍モデル・脳腫瘍モデルに小型加速器中性子線源による照射を行い、治療効果を検討する。また、ホウ素取り込み改善に関する基礎研究を行い、将来臨床応用可能な新規ホウ素化合物の開発を目指す。
②BNCT 臨床研究:AMED臨床試験を目指して、これまでの加速器ベースで新規膠芽腫のBNCTを行った研究はなく、First in Human studyとなる。初回にBNCTを行い、その後にX線分割照射をMRIのT2high intensityに加えることにより、これまでに安全性の担保、播種や遠隔再発を防ぐことが原子炉を用いて実証されており、これを加速器にて検証していく。
③臨床試験画像誘導システムによる治療装置のシュミレーションを行う。
