研究成果の概要

ChemProteoBaseの高度化を目的に、既存薬剤のプロテオームデータを取得してデータベースを拡充するとともに、パスウェイ解析やキーワード解析ができる新たなツールを開発した。また、ChemProteoBaseのシステム・データベースを活用して、2次元電気泳動法とセルサーマルシフトアッセイを組み合わせた新たな薬剤標的同定直接法2DE-CETSAを開発した。がん細胞の代謝機構を標的とした生物活性リガンドのスクリーニングを実施し、NPD10084がPKM2を標的とすることを明らかにした。そのほか、新規ながん代謝阻害化合物を見出し、標的を明らかにするとともに、天然から多数の新規活性物質を見出した。

研究成果の学術的意義や社会的意義

有用な生物活性リガンドが利用されるためには、その標的分子が明らかであることが必要である。これらには表現型スクリーニングで得られた化合物も多く、酵素阻害剤などのように目的とする分子が想定された化合物とは異なり、標的分子を同定することは必ずしも容易ではない。薬剤標的を直接法（2DE-CETSA）と間接法（ChemProteoBase）の双方を用いてプロテオームワイドに解析できる独自のケモプロテオミクス基盤は、生物活性リガンドの標的同定を加速させるとともに、本プロジェクトで見出された新規生物活性物質は、今後バイオプローブや医薬品等の開発に貢献することが期待される。
