研究概要

マウス初期胚に存在する多能性細胞から、始原生殖細胞,精原細胞、卵母細胞に至る一連の細胞系譜の発生過程を、全遺伝子発現プロファイル、DNA メチル化プロファイル、および細胞核構造の変化やX染色体活性変化を指標として包括的に捉え直し、各発生ステージで機能する遺伝子群を網羅するとともに、それぞれの指標が大規模な変動を示すステップ、すなわち発生プログラムの転換点を同定した。また、この細胞系譜で働く遺伝子のエピジェネティック制御に関与すると考えられる生殖細胞特異的なDNA 低メチル化領域を発見した。発生プログラム転換に寄与する遺伝子の同定、機能解析を変異マウス等を利用して行い、Vps52 遺伝子が多能性胚細胞から原始外胚葉への転換に必須の遺伝子であることを突き止めた。以上の研究遂行に必要な新規技術の開発、幹細胞および遺伝子改変マウス等の研究リソースの開発を行ない、さらに、これらの技術、研究リソースを活用した領域内の有機的連携研究を実施した。
