研究概要

シンデカン-4のノックアウトマウスの解析から、シンデカン-4上のヘパラン硫酸プロテオグリカン鎖は胎盤での血液凝固の防止と腎臓での生体防御に役立つことが判明した。ヘパリン結合性の成長因子ミッドカインの受容体の1つはコンドロイチン硫酸プロテオグリカンである受容体型プロテインホスファターゼ(PTPζ)であるが、ミッドカインが識別するコンドロイチン硫酸鎖はE鎖でしかもデルマタン硫酸ドメインをもつことがコンドロイチン硫酸Eとの反応性およびコンドロイチナーゼB消化の結果判明した。ミッドカインとPTPζの識別は神経細胞の移動のみならずその生存、そして骨芽細胞の移動においても重要であった。ホルモン糖タンパク質の糖鎖合成に関与するN-アセチルガラクトサミン-4-スルホトランスフェラーゼをcDNAクローニングした。従来のN-アセチルグルコサミン-6-スルホトランスフェラーゼとは基質特異性の異なるN-アセチルグルコサミン-6-スルホトランスフェラーゼをcDNAクローニングした。ヒトのEXTのホモローグである線虫のrib-2遺伝子はヘパラン硫酸の糖鎖合成の開始と伸長に関与する新しいα-1、4-Nアセチルガラクトサミニルトランスフェラーゼであることを明らかにした。コンドロイチン硫酸E(コンドロイチン-4、6-ジ硫酸)の合成を司るスルホトランスフェラーゼを用いて硫酸化度の異なるコンドロイチン硫酸Eを作り、その機能解析の基盤を作った。ブタからヒトへの異種移植の拒絶において最も大きな障害となるα-ガラクトース抗原を除去するエンド-β-ガラクトシダーゼCを発現させ、精製した。精製酵素をブタに静脈注射すると、一時的にではあるが、α-ガラタトース抗原を除去できた。
