研究概要

Okamuraによって発見されたIL-18はIL-12と協同しTh2誘導を抑制しTh1型サイトカイン(IFN-Y)産生及びTh1型細胞への分化を強力に誘導すると考えられていた。申請者は新しいNK細胞ポピュレーションIL-13産生NK細胞を発見し報告した(JI,Jan 1,1999)。その上申請者はIL-18がIL-2と共同しTh2サイトカインであるIL-13をNK細胞及びT細胞から誘導されることを初めて報告した(JI,1999)。またその後申請者と中西らは別々にIL-18がIL-4,CD40 ligand(CD154)を介しIgE誘導に関与していると報告した(EJI,2000,Nature Immunol,2000)。以上よりIL-18はTh1誘導だけでなくIL-13産生NK細胞及びTh2T細胞誘導を介し抗体(IgE)産生誘導、Th2誘導、アレルギー成立にも関与していることが示唆された。
申請者はIgH(B cell)promoter(pEμIgH)及びkeratinocyte promoter(K5)を用いtissue-specific expression IL-18 transgenic(Tg)mouseの樹立した。これらのIL-18tgマウスを解析中したところある。IpEμIgH-IL-18TgマウスはTh2 response,K5-IL-18TgマウスはTh1 responseの傾向を示した(一部論文投稿中及び現在解析中)。IL-18がTh1/Th2の両方の誘導に関与していることが証明された。
