研究成果の概要

細胞機能は環境に応じて変化する。各種細胞膜蛋白質の細胞外ドメインが細胞に特異的な生理機能を担う。血小板ではGPIIb/IIIaの細胞外ドメインの高次構造変化が細胞活性化後に起こる。活性型構造のGPIIb/IIIaはフィブリノーゲンなどのリガンドに結合し、血小板凝集、血栓の成長に寄与する。活性化に伴う血小板細胞内の環境変化はカルシウムイオン濃度上昇など大きくない。小さな細胞内環境変化が細胞外ドメインの大きな構造変化を惹起する力学メカニズムとして「分子レバレッジ仮説」を提案し、妥当性を分子動力学計算により検証した。

研究成果の学術的意義や社会的意義

血小板細胞は刺激に応じて活性化する。非活性化血小板は凝集し、活性化血小板は凝集する。血小板凝集は血小板膜糖蛋白GPIIb/IIIaの活性化構造変化に依存する。活性化とともに血小板細胞内のカルシウムイオン濃度は上昇する。GPIIb/IIIaの細胞内ドメインにはTalinなどの接着斑蛋白が結合する。細胞内ドメインの構造も細胞活性化により変化すると想定されるが細胞内ドメインは短いので、構造変化は大きくない。この小さな細胞内ドメインの構造変化が、細胞外ドメインの大きな構造変化と血小板細胞の性質変化を惹起するメカニズムは未知である。このメカニズムとして分子レバレッジ仮説を提案し、妥当性を検証する。
