研究概要

2つのポリケチド抗生物質ランカサイジン(LC)・ランカマイシン(LM)を生産する放線菌Streptomyces rochei 7434AN4株の線状プラスミドpSLA2-Lの機能解析を行った。LCの大員環形成が、LkcEによるC-18位アミノ基酸化により生じることを明らかにした。また、LM生合成における水酸化、糖付加の順序を解明した。γ-ブチロラクトン(GB)を鍵物質としたLC・LM生産制御カスケードの解析も行い、GB生合成遺伝子srrX→GBリセプターsrrA+┥SARP型活性化因子srrY→LC, LMというカスケードを明らかにした。
