研究概要

機能未知のイノシトールリン脂質PI(3,4)P2を脱リン酸化して分解する酵素には、2 つのアイソザイム(L-PIPase とP-PIPase)が存在する。本研究では、L-PIPaseとP-PIPase の各々が特異的に担う生理機能を明らかにし、この特異性の分子メカニズムを、各アイソザイムの基質となるPI(3,4)P2の分子種の違いに求めて詳解することを目指した。その結果、L-PIPaseは神経細胞死に、P-PIPaseは発癌に特異的に関与することを見出した。また、L-PIPaseとP-PIPaseが種々のPI(3,4)P2分子種に対して異なる活性を有することを見出した。以上の結果より、両分子の生理機能の差が、基質とするPI(3,4)P2分子種の違いに起因する可能性が示唆された。
