研究概要

ラクダ科動物のH鎖抗体の特徴を生かし、インフルエンザウイルスHA蛋白の開裂を特異的に防ぐことで、ウイルス感染症の治療研究を意図した。血清でのHI試験では、熱処理によるタンパク変性により抗原認識能が顕著に低下するが、免疫後期においては僅かながら熱耐性の抗体分子が産生されていた。精製したH 鎖抗体のIgG2とIgG3は、熱処理後でも1,440nmol、1,400nmolの濃度でHI(+)であった。VHH抗体遺伝子特異的PCR産物をファージミドベクターに導入し、大腸菌DH5αに形質転換したクローンを分離した。より強いアフィニティーを持つVHHクローンを単離するため、人為的な変異導入やDNaseI処理による遺伝子の再構築の可能性についても検討する必要がある。
