研究概要

EGFR-TKIのひとつであるエルロチニブの感受性を決定するEGFR遺伝子変異以外の要因を明らかとし、同時に腫瘍の悪性形質におけるEGFRの下流シグナル系の役割を検討し、同経路の遺伝的個人差の存在の有無を検討とすることを目的とした。野生型EGFR遺伝子をもつ非小細胞肺癌症例を対象に、エルロチニブを投与する臨床試験を行い、cDNAマイクロアレイ、RT-PCRによるバイオマーカー検索を行った。非小細胞肺癌細胞株、中皮腫細胞株を用いてEGFR下流シグナル系、薬剤相互作用、耐性機構のための基礎的検討を行った。Thymidylatesynthaseの発現に注目して解析を加えたが、中皮腫細胞株においてはpemetrexed耐性機構に関連はなく、非小細胞肺癌細胞株においては、メトホルミンによる制癌効果を認める結果を得、特に腫瘍形成初期における制癌効果が高いことを明らかとした。
