研究概要

本研究の目的はRNAiスクリーニングを用いて、骨髄異形成症候群(MDS)の病態進展・治療抵抗性機序を網羅的に解析することである。
本研究で我々はヒト遺伝子に対するプール型shRNAプラスミドライブラリーであるCellecta社のDECIPHERをMDS由来細胞株に導入し、Azacitidineで30日間処理した。処理後の細胞のDNAを次世代シークエンサーでシークエンスし、各shRNAコンストラクトに特異的なバーコード配列の頻度をAzacitidine処理群とControl群で比較した。その機能喪失によりAzacitidine耐性が誘導される候補遺伝子として158個の遺伝子を抽出することができた。
