研究概要	本年度は、とくに、自己の癌細胞や自己組織に発現したheat shock protein(HSP)、あるいは、アロのMHCークラスI抗原などを認識して、これに対して傷害活性を示すgamma delta T細胞の誘導とその機構について研究を重ね、以下のような多くの研究成果を得た。 (1)in vitroでのgamma delta T細胞の誘導刺激を検討し、その結果、実験室で継代を重ねているヒト癌細胞(K562)、マイトマイシンCでDNA合成を抑性した自己のマクロファージ、あるいはアジューバント活性の強いグラム陽性細菌(加熱死菌)などに、ヒト抹梢血リンパ球との培養によって著るしい細胞傷害活性をもったgamma delta T細胞を誘導する活性のあること、(2)これらのgamma delta T細胞の誘導におけるDNA合成はILー2非依存性であるが、ILー3とILー4の産生を伴うこと、反応に自家血清を必要とするとこなどを明らかにした。(3)さらに、誘導因子に関して最もよく研究したのはグラム陽性球菌(Streptococcus pyogenes)であるが、この菌体の誘導活性因子は細胞壁やその構成々分(peptidoglycan)にはなく、もう1つの生体膜である細胞質膜にあること、その活性因子はlipid bilayerにくみことまれている分子量10kDaの蛋白であることなどを明らかにした。gamma delta T細胞の誘導機構のモデルとして、現在、この活性単位を分離し、その構造を決定中である。この成績を下に、癌細胞やマクロファージに発現しているgamma delta T細胞を刺激誘導する因子の性状を明らかにして行きたい。
