研究概要	ラット繊維芽細胞WFBは、血小板由来成長因子(PDGF)、バソプレシン、ボンベシンおよびPGF_<2α>の刺激に応答し、細胞内遊離Ca^<2++>濃度([Ca^<2+>]i)の上昇とホスファチジルイノシト-ル4,5ビスリン酸(PIP_2)のホスホリパ-ゼC(PLC)による加水分解を起こす。活性化C-Ha-ras遺伝子でトランスフォ-ムしたWFB細胞では、PDGF刺激に対する[Ca^<2+>]i上昇応答が特異的に消失した。この消失は、ras癌遺伝子による癌化で特異的見られる現象であり、ポリオ-マミドルTにより癌化したWFBでは消失しない。Ras癌遺伝子のコ-ドするras p21タンパク質がどのような機構によりPDGF受容体を介するPLCの活性化に干渉するのかを明らかにする目的で、精製したras p21タンパク質をWFB細胞に導入する実験を行った。12番目にバリンを有する活性化C-Ha-ras p21タンパク質は、合成遺伝子の発現ベクタ-pHR-L9を大腸菌に発現させたものから精製した。ras p21タンパク質をWFB細胞に加え、scrape-loadingにより細胞にras p21タンパク質を封入した。ras p21タンパク質を封入すると、細胞の接着性が増大し、20時間保温する間に細胞数も増加した。それに伴い、PDGF刺激によるイノシト-ルリン酸の生成応答が、牛血清アルブミンを封入した対照細胞に比較して小さくなった。このようなrasタンパク質の効果は、C端8コのアミノ酸が欠落したras p20タンパク質では認められなかった。ras p21タンパク質をscrape-loadingした後、3時間後に膜画分を調整しイノシト-ル脂質の分解を検討した所、グリセロホスホイノシト-ル生成の亢進が認められた。ストレプトリジンO処理によりタンパク質に対し透過性を持たせたWFB細胞を用いて、PDGF刺激及びバソプレシン刺激に伴うイノシト-ルリン酸生成応答に対するras p21タンパク質の効果を検討したが、効果は認められなかった。
