研究概要

アンドロゲン依存性癌細胞(SCー3細胞)増殖の分子機構を検討した。アンドロゲンにより誘導される増殖因子は、抗bFGF抗体で活性が中和され、FGF受容体と結合するが、ヘパリンにより失活される点でFGFとは異なる分子であることが判明した。Conditioned Mediumをヘパリンカラム、カッパ-カラム及びC_4カラム(逆相HPLC)にかけることにより精製をすすめた。又アンドロゲン刺激下のSCー3細胞から作製したExpression cDNA LibraryをNIH3T3細胞にTransfectionし、形態変化(Transformation)と自己増殖能を獲得したコロニ-を単離した。このコロニ-に挿入されたcDNAをシ-クエンスしつつある。このライブラリ-から、FGF受容体をコ-ドするcDNAを単離し、COSー7細胞で発現可能であることを確認した。このcDNAをシ-クエンスすると、マウス脳で発現しているFGF受容体と比較し、N端近くに12アミノ酸が附加された832個のアミノ酸よりなる蛋白であることが判明した。tyrosine kinase領域には、脳のそれと全く同一のシ-クエンスであった。しかし、アンドロゲンやbFGFでSCー3細胞を刺激すると、FGF受容体のmRNAレベルは著増した。NIH3T3細胞では、bFGF刺激によりダウンレギュレ-ションを受けた。SCー3細胞はFGF関連増殖因子の刺激を増幅する機構が存在し、非癌細胞での制御とは異なることが判明した。
エストロゲン依存性癌細胞(Bー1F細胞)は、ロイコトルエンC_4,D_4やE_4で抑制された。又Bー1F細胞にはロイコトルエンレセプタ-D_4受容体が存在した。Bー1F細胞をエストロゲンで刺激すると、ロイコトルエンC_4,D_4やE_4の産生が低下した。エストロゲンの作用点を明らかにする為に、Bー1F細胞の5ーリポキシゲナ-ゼを測定すると、エストロゲン刺激で低下することが明らかとなった。エストロゲン依存性癌増殖は、5'ーリポキシゲナ-ゼを低下させることにより生じることが判明した。
