研究概要

MPTPがヒトでパ-キンソニズムを発症させることが明らかとなって以来、パ-キンソン病が少なくとも一部外因毒物により発症する可能性が高まっている。このことから、MPTPに類似の構造を有し、食物等に含まれるテトラヒドロイソキノリン(TIQ)が候補物質との仮説にもとづき、TIQ(20mg/kg)を最長104日間リスザルに投与(皮下注)した。その結果、行動上の異常を引きおこすと共に、通常臨床的に抗パ-キンソン薬として使用されるLーDOPAを投与すると病状の改善をみた。一方脳の生化学的分析の結果、線条体でのド-パミン、チロシン水酸化酵素、総ビオプテリンなど、パ-キンソン病で著減する諸物質がTIQ投与サルでも減少していた。しかしながら、最も重要な所見である黒質ド-パミンニュ-ロンの脱落はこれまでの所、形態的につかまえることが出来ていない。ただ、チロシン水酸化酵素活性を免疫組織化学的に染めると、同陽性細胞は、コントロ-ルの約半分以下に減少していた。なお、TIQと関連しているNーメチルーTIQも、約100日間、老化サル(赤毛)に皮下注を試ろみたが、この場合には、一過性の急性にみられる行動異常が注射直後にみとめられたのみで、明らかな慢性的変化は認めなかった。
