海産有用生理活性物質の探査と利用に関する研究

研究概要	微生物、藻類、無脊椎動物を含む広範な海洋生物について生理活性物質の探査、構造決定、作用解明が行われた。ヒラメ飼育水槽由来Vibrio菌156株からは24株(15.3%)に抗魚病ウィルス活性を検出した。更に、Pseudomonas sp.48HSー27株の抗ウィルス成分を精製し、アミノ糖の可能性を指摘した(絵面)。90種の微細藻をスクリ-ニングし、5種の藍藻及び、渦鞭毛藻とラフィド藻各一種に、きょく皮動物受精卵の卵割阻害活性を検出した。有望種4種の大量培養と成分の単離が進めている(山口)。また、2種の渦鞭毛藻の生産する強力な抗カビ成分amphidinolとgambieric acidsの構造が決された(安元)。大型海藻のコメノリからは2種の免疫賦活物質が単離・精製され、化学性状と作用の検討の結果、陸上植物由来のマイトゲンとの異同が明らかにされた(堀)。腹足類のアメフラシから得られた抗腫瘍タンパク質のアプリシアニンを腫瘍細胞培養液に添加すると、2時間以内に高分子合成を阻止し、10数時間後に細胞溶解を引き起こした。細菌に対する溶菌作用はないが、短時間にタンパク質や核酸などの高分子合成を阻害した。現在、各種の観点から作用機構を追究している(神谷)。刺毒の生化学試薬としての開発を目指して、オニヒトデのホスホリパ-ゼAの一成分を精製・単離し、分子量14000の酸性タンパク質と推定した。オニダルマオコゼ刺毒から単離・精製された致死因子も、分子量 90000の酸性タンパク質であった。目下、作用の解明を行っている(塩見)。海綿を中心とする無脊椎動物の有用物質を探査し、単離した成分の構造決定を行った。八丈島産のTheonella属海綿からは特異な環状ペプチドのorbiculamide A、テトラミン産配糖体のaurantoside AとB、及びmycalamideA誘導体が細胞毒性物質として得られた。特に、mycalamide誘導体の著しい高腫瘍活性が注目された(伏谷)。
