研究概要

低エネルギー放射線は放射線の生物作用の基礎過程の解明の強力な武器となるとされているが、従来より適当な線源がないことから生物実験のデータがなく、放射線の生物作用の理論的解明や人体影響の評価の障害となっていた。本研究は、最近利用可能となってきた単色軟X線やエネルギーを制御された低エネルギー中性子を駆使し、生物効果の定量解析から放射線の生物作用の基礎過程の解明を目指してきた。プラスミドDNAの分子解離は、乾燥系では構成原子のエネルギー吸収で説明できるが、水溶液系では二次電子の寄与が重要であることが分かった。このことは、イーストの突然変異やマウス培養細胞の染色体突然変異でもリン原子の内殻電離を利用した実験からも実証された。さらに、低エネルギーX線の場合、分子濃度と共に効果が上昇することは光電吸収によるオージェ電子の寄与が増えることを意味する。X線造影剤の沃素を標的とした実験から沃素原子の共鳴吸収が染色体異常を上昇させることが証明され、がん治療としての光子活性化療法の可能性を示す。1MeV前後の高速中性子は細胞死、突然変異、がん化に関して高い生物効果を示すが、中性子フィルターを使って20keV前後にまで減速しても生物効果は変わらないことが明らかとなった。この結果は、低エネルギー中性子の放射線荷重係数の再検討の提唱につながる。事実、原爆被爆者での突然変異頻度の線量効果関係は広島と長崎とで異なり、広島原爆における低エネルギー中性子の生物効果を無視できないことを裏付ける結果も得られた。わが国におけるエネルギーを制御した低エネルギー領域のX線・中性子の生物照射場の開発は世界に一歩先んじており、得られた成果にはこの分野における先導的なものが多い。
