精子運動調節機構の分子生物学的研究

研究概要

今年度は、サケ科魚類の精子運動開始の引金を引く分子量15,000のタンパク質(15Kタンパク)は分子量1000,000のタンパク質(1Mタンパク)の構成成分であることを明らかにした。更に、1Mタンパク質のモノクローン抗体の作成を行い、64種の抗体を得、各々のニジマス精子除膜モデルの運動に対する影響について調べた。その結果、13種について阻害活性が認められた。現在、活性の強い数種類について1Ggを精製中である。また、15Kタンパク質単体についてもモノクローン抗体作成を行っている。今年度は新たに、精子運動調節における多機能型高分子量プロテアーゼ(プロテアソーム)の役割について調べ、まず、ウニ精子からキモトリプシン様活性を持つ分子量650KDa,950KDaのプロテアーゼを精製し、そのうち650KDaプロテアーゼがプロテアソームであることを明らかにした。更に、シロサケにおいても同様のプロテアーゼおよびプロテアソームが存在し、それらがATP濃度依存的に精子運動を調節していることを明らかにした。海産魚の精子運動開始については、昨年度のCa^<2+>,pHの研究に加え、K^+の影響、膜電位の変化についてイオノフォア,蛍光指示薬を用いて調べ、細胞内Ca^<2+>およびK^+が精子運動開始に最も重要であり、細胞内pH上昇が補足的な役割を果たしていること、膜電位変化はイオン,pHの変動の結果として生じる可能性が強いことが明らかとなった。ニシン精子の運動活性化については、卵由来の活性化物質の精製が完了し、そのモノクローナル抗体を作成した。現在、その精子運動活性化に与える影響について検定中である。また、cDNAクローニングについても研究を続行中である。シロサケでは、卵の卵門付近に精子を誘引する作用のあることが水浸レンズを用いた観察で明らかとなった。ホヤ卵由来の精子走化性物質に関しては、その性質が昨年度様々に調べられたが、今年はその精製がほぼ完了の域まで達した。
