研究概要

◆精子運動開始機構に関しては、海産魚及び淡水魚で外部浸透圧を繰り返し増減または滅増することにより精子の運動の開始・停止が繰り返し起こるという興味深い発見がなされ、また、界面活性剤で細胞膜を除去した除膜モデルの精子では外部のK^+イオン濃度の増減・滅増で鞭毛軸糸の運動の開始・停止が繰り返し起こることも見い出された。更に、除膜精子は海産魚・淡水魚の両方で活性化溶液中のpHを増減することによって、やはり運動の開始・停止が繰り返し起こることも明らかになった。以上の結果は、魚類精子運動は外部浸透圧の変化によって細胞内K^+濃度及びpHの増減が起こり、この両要素が鞭毛軸糸の運動装置を制御していることを示している。◆精子運動活性化については、ニシン卵由来精子活性化物質(HSAPs)の遺伝子がクローニングされ、全アミノ酸配列からトリプシンインヒビターに類似していることが明らかにされたが、今回ニシン精子にそのリセプターの候補としてトリプシンが数種類あることが明らかとなった。更に、市販のトリプシンインヒビターがHSAPsと同様に精子活性化能のあることも明らかとなり、トリプシンインヒビターとトリプシンの結合が精子運動活性化の引き金となるという考え方が提示された。◆精子活性化・走化性物質に関してはユウレイボヤ卵海水からほぼ精製を完了していたが、今回は質量分析法を用い分子量288または1,089の比較的低分子物質であることが分かってきた。また、精製標品が精子活性化・走化性の両方の作用を持っていることから、1個のタンパク質が2つの機能を果たしているという興味深い結果も得られた。また、様々なCa^<2+>チャネル阻害剤を用いた実験から精子活性化に働くことが分かっていたCa^<2+>は、T型Ca^<2+>チャネルを通過し細胞内へ活性化情報を伝達することが明らかとなった。◆精子運動性の調節に関しては精子内の多機能型プロテアーゼの存在とその役割についての研究が開始された。
