研究概要

自由行動動物における環境適応行動の長期間多元的解析システムの開発と応用を目的とし、平成5年度は以下の成果を得た。1.無麻酔・自由行動下動物における脳マイクロダイアリシス解析の実用化:前年度までに開発した脳マイクロダイアリシス装置を用い、自由行動下ラットの内側前頭前野におけるノルアドレナリン(NA)動態を解析した。(1)保持時間が標準物質(NA)と一致するクロマトピークが(a)高K^+液、methamphetamineまたはdesipramineをプローブ中に潅流すると増大する、(b)TTXまたは無Ca^<++>リンガー液の潅流、或いはalpha-methyl-p-tyrosine腹腔内投与により減少する、(c)背側前脳束刺激または切断でそれぞれ増大または消失する事などから、延髄青斑核NA含有ニューロンから投射する前頭前野の神経終末から放出されたNAを測定していることを確認した。(2)拘束ストレス(1hr)により、NA放出は持続的に176%増加し、この増加はdiazepam(抗不安薬)により抑制された。(3)FG7142(不安誘起物質)投与によりNA放出が増加し、それはflumazenilにより抑制された。2.脾臓マイクロダイアリシス解析の実用化:前年度までに完成したシステムを用い、無麻酔ラットにおいて、脾臓交感神経終末からのNA放出を解析した。安静時のNAレベルは14.0pg/40mu1であり、拘束ストレスで6-7倍に達し脾神経切断で消失する。IL-1beta腹腔内投与により脾NA放出は増加し、これは脳CRFニューロンの活性化により誘導されることを確認した。3.テレメーターによる体温、心電図及び運動レベルの記録システムの実用化:送信器を腹腔内埋め込みし、体温、心電図、運動レベル、摂食量、飲水量を長期記録し、各パラメーターの基礎24時間リズムデータを観察した後、IL-1 receptor antagonist投与の効果を2-3日間にわたって検索し、十分実用に耐えることを確認した。
