蛋白質燐酸化酵素及び脱燐酸化酵素サブタイプ特異的発現細胞系の確立とその有効利用

研究概要	蛋白質燐酸化酵素と脱燐酸化酵素は細胞内情報伝達の中枢をなす機構として、注目を集めている。最近の遺伝子工学の進歩により、従来は単一の分子種からなると考えられていたこれらの酵素がいくつかのサブタイプを持つことが明らかにされてきた。本研究では、プロテインキナーゼAやプロテインキナーゼC等のタンパク質燐酸化酵素、カルシニューリンなどの蛋白質脱燐酸化酵素各サブタイプの生理的役割を明らかにするため、各サブタイプを過剰生産する細胞系を確立することを目的とし、以下の結果を得た。(1)カルシニューリンの触媒サブユニットであるAサブユニットの2つのサブタイプAαとAβのcDNAを発現ベクターに組み込み、AαとAβそれぞれを特異的に大量発現する大腸菌株を樹立した。発現された蛋白質を抗原として、Aα、Aβに対する特異抗体を作製した。これらの抗体を用いて、ラット脳におけるこれら2つのサブユニットの分布を調べた。また、免疫沈降法により、牛脳から精製したカルシニューリンをAαからなるヘテロダイマーとAβからなるヘテロダイマーに分離し、FK506/FKBP複合体の効果を調べた。(2)精巣特異的に発現する新しいカルシニューリンBサブタイプのcDNAクローニングに成功し、カルシニューリンBには異なる遺伝子によってコードされる2つのサブタイプが存在する事を明らかにした。(3)ラットプロテンキナーゼAの触媒サブユニットの2つのサブタイプCα、Cβをクローニングし、ラット各臓器、脳内分布及び培養細胞における分布を明かにした。(4)Cキナーゼサブタイプの線条体黒質系における分布を明らかにした。(5)発達期脳におけるプロテインキナーゼC分子種の変化を調べ、分子種によってその出現時期が異なる事を明らかにした。(6)GABA_B受容体とプロテインキナーゼCをアフリカ爪ガエル卵に発現させ、GABA_B受容体を介する電気現象は、プロテインキナーゼCにより制御される事を示した。
