急性白血病でみられる染色体転座部位に座位する癌関連遺伝子の研究

研究概要	我々は、11q23を含む複数の染色体に異常を持つ乳児急性単球性白血病細胞を用いてMLL遺伝子の転座相手遺伝子のクローニングを行った。末梢血白血病細胞よりcDNAライブラリーを作成し、切断点を含むMLLcDNAの0.9kb BamHI断片をプローブとしてスクリーニングを行った。その結果得られた2個のキメラクローンは、一方は5'側にエクソン6までのMLLcDNAと3'側にt(6;11)転座の6番の遺伝子として単離されたAF6が、もう一方は5'側に100bpの未知の塩基配列(AF-T)と、3'側にエクソン7より下流のMLLcDNAが融合していた。AF-Tはメッセージの大きさが約2kbで少なくとも269のアモノ酸をコードしていると考えられる。現在までにクローニングされている8つのMLL遺伝子の転座相手遺伝子とは今のところ共通点は見い出せず、また特別なモチーフも見つかっていない。Northern blotによる解析では、myeloid系、lymphoid系の白血病細胞株でひろく発現がみられ、またヒトの胎児組織でひろく発現していた。特に胎児期の造血の中心である肝組織での発現が強く、この遺伝子が乳児の白血病細胞からクローニングされたものであることと合わせ、血球系の未分化な段階で働く遺伝子である可能性が示唆される。現在さらにAF-Tの機能の解析を進めると共に、少なくとも3つの染色体が同時に転座を起こしていると考えられるこの細胞の持つ複雑な染色体転座について、FISH,genomeのクローニングなどを行い詳細な解析を進めている。
