研究概要	重金属の毒性発現における感受性修飾因子として、生体の活性酸素除去能力が関与しているかどうかを検討するため、glutathione peroxidase,CuZn-SOD,Mn-SOD,catalaseの活性を遺伝子導入法により上昇させた細胞系(HeLa細胞)を用い、様々な重金属塩及びその錯体、あるいは紫外線照射による毒性発現の変化を調べた。その結果、catalaseとCuZn-SODの両cDNAを導入した細胞が、パラコートや白金錯体であるシスプラチン、あるいは紫外線(UV-B)の大量照射に対して耐性になっていることがわかった(井村)。環境化学物質に対する感受性を支配する遺伝的な要因として、チトクロームP450の分子種であるCYP2D6に注目し、日本人におけるこれらの酵素の遺伝的変異・多型について検討した。前年度までの研究を見いだした新たな変異、及びこれまでに報告されている遺伝子変異について、日本人における頻度を検討した結果、約0.2%にこれらの遺伝子変異が認められた(鎌滝)。日本人において鼻アレルギーや気管支喘息の誘因となる主要アレルゲンである家ダニ(コナヒョウヒダニ)の抗原(Der fI)に対する免疫応答の個体差の機構を解明するため、HLAクラスII遺伝子の異なる5人のダニアレルギー患者から、Der fIペプチドに特異的に増殖性応答を示すT細胞株を樹立した。これを用い、T細胞が認識する抗原ペプチドのエピトープを13種類同定した(西村)。電磁場に対する感受性因子が、放射線感受性因子と同一のものであるかどうかを検討するため、プラスミドベクターを用いたDNA鎖切断修復率を調べた。その結果、放射線感受性細胞株も、非感受性株も、電磁波への曝露により鎖切断の再結合の上昇が見られたことから、電磁波に対する感受性因子と放射線感受性因子が異なっていることが示唆された。(巽)。
