高DNA修復能トランスジェニックマウスでの発癌、突然変異、老化の研究

研究概要	本研究ではDNA修復機構とがん発生予防との関連性についてDNA損傷修復に関わる遺伝子導入動物を用い以下の研究を行った。 1)我々のグループが作製した大腸菌由来06-メチルグアニンDNA メチルトランスフェラーゼ(メチルトランスフェラーゼ)遺伝子導入マウス(adaマウス)はメチルトランスフェラーゼ遺伝子を高発現しており、通常同系マウスの約3倍程度である。このadaマウスを用い、アルキル化肝発癌剤による肝腫瘍誘発実験を行い、メチルトランスフェラーゼが誘発腫瘍を予防していることをすでに証明した。今年度はこの系統のマウスで生後1年以後肝腫瘍が雄で高頻度に自然発生することが知られているのでadaマウスと同系対照マウスを2年間観察し、その自然発生腫瘍について検討を行った。その結果、自然発生腫瘍発生率は両群共48%と差は認められなかった。しかし、病理組織学的検索の結果、adaマウスでの悪性腫瘍の割合は対照マウスに比べ有意に低く、メチルトランスフェラーゼが腫瘍の悪性化を抑えていることが示唆された。
