高DNA修復能トランスジェニックマウスでの発癌、突然変異、老化の研究

研究概要

大腸菌由来O^6-メチルグアニンDNA メチルトランスフェラーゼ(MGMT)遺伝子を導入し高発現するトランスジェニックマウス(adaマウス)を作製し、ニトロサミンによる肝腫瘍誘発実験を行った結果、MGMTが誘発発癌を予防している事をすでに証明した。今回この系統のマウスは生後1年以後、肝腫瘍が雄で高頻度に自然発生をすることが知られているのでMGMT活性が肝等で正常マウスに比べ約3倍上昇しているadaマウスと対照マウスを2年間観察しその生存曲線及び自然発生肝腫瘍について検討を行った。実験はadaマウス雄;130匹、雌;62匹.対照C3H/HeNマウス、雄;48匹、雌;14匹を用いた。自然発生肝腫瘍の発生率は良性、悪性の腫瘍をあわせると両群共48%と差は認められなかった。しかし、病理組織検索の結果adaマウスでは腺腫が60%、肝癌が28%であったのに対し、対照マウスでは腺腫が39%、肝癌が60%で対照マウスではadaマウスに比べ悪性腫瘍の占める割合が有意に高かった。MGMTの高発現が腺腫から肝癌への悪性化に関与している可能性が示唆された。
