胚中心B細胞の発生・分化機構と異常に関する研究

研究概要	B細胞の生存、活性化、免疫記憶に重要な役割を果たすと考えられている胚中心に関しては不明な点が多い。本研究では胚中心におけるB細胞の増殖・分化、クラススイッチおよび超変異(hypermutation)の分子機構を明らかにし、それを撹乱する要因を明らかにするとともに、その制御法の開発を目的としている。 (研究方法と研究成果)(1)B細胞の生存延長や活性化を誘導するIL-5、IL-4、抗CD38および抗CD40抗体刺激に注目し、これらの胚中心B細胞に対する作用を解析した。その結果、胚中心B細胞の一部がIL-5ないしCD38を発現しており両分子をともに発現しているB細胞も検出された。また、胚中心B細胞のほとんどがIL-4RとCD40分子をともに発現していることを明らかにした。(2)B220^+/IgD^-/PNA^+を指標として胚中心B細胞を濃縮し、IL-4、IL-5、抗CD40抗体刺激に対する応答性を解析した結果、これらの刺激が共存した場合に最も効率よく胚中心B細胞を抗原特異的IgGl抗体産生細胞へと分化させること、IL-4と抗CD40抗体刺激により胚中心B細胞の表面のIL-5Rの発現が増強されることを明らかにした。(3)脾臓B細胞を抗CD38抗体で刺激すると有意な増殖反応が見られたがIg産生は見られなかった。その系にIL-5Rを共存させると増殖反応は10倍以上増加し、抗体産生細胞への分化の指標と考えられているBlimp-l遺伝子の発現とIg産生が誘導された。またCD38刺激にともないBruton′s tyrosine kinase(BtK)のチロシンリン酸化が見られ、Btk遺伝子に変異のあるXIDマウスのB細胞はCD38刺激に応答しないことが初めてわかった。(4)IL-5Rα鎖はIL-5との特異的な結合に必須であるのみならず、シグナル伝達にも必須である。α鎖の細胞内ドメインのプロリン残基に富む領域(Pro-Pro-X-Pro)がシグナル伝達(増殖、チロシン残基のリン酸化、proto-onco geneの発現)に必須であること、3個のProのうち1個の存在がJAK2キナーゼの活性化とリン酸とSTAT5の活性化に必須であることを明らかにした。
