研究概要	1)SLE病態の多様性を規定している素因遺伝子を明らかにするために、SLEのモデルマウス系、NZBxNZW(B/W)F1,NZWxBXSB(W/BX)F1,NZBxBXSB(B/BX)F1で先ず病態の比較をした。その結果、抗DNA抗体を伴うループス腎炎は全ての系に発症したが、一方、血小板減少症はB/BXF1に高度に、W/BXF1に軽度に認められたが、B/WF1には見られなかった。抗リン脂質抗体症候群はW/BXF1にのみ発症した。 2)退交配系によるmicrosatellite DNA解析及びH-2-congenic系による解析で、SLEには、共通した素因遺伝子と病態の多様性を支配する複数の遺伝子が存在することが解った。特に、NZBとBXSBの第4染色体尾部に、自己反応性B細胞の異常な分化に関わる、SLEに共通する素因遺伝子が存在する事を明らかにした。この遺伝子はhyper‐IgMの発症に関与するので、Imh‐1と命名した。 3)W/BXF1系にユニークな抗リン脂質抗体症候群は、H-2ハプロタイプを代えると発症が著しく低下するので、この系に特有のH‐2z/bヘテロ接合性に拘束されていることが明かとなった。 4)NZW.H‐2^<d/d>‐congenic系に、クラスII遺伝子Aβ^zをtransgene‐congenicにしたマウスの樹立に成功した。現在、NZB(H‐2^d)との交配F1を生産し、抗DNA抗体の産生に関与するクラスIIA分子の役割を検討しているが、予備試験でAα^d/Aβ^zが重要な拘束分子であることが示唆されている。
