グリオーマ運動性因子による運動制御の解析-腫瘍浸潤の分子機構解明と治療法の開発

研究概要

本年度は、グリオーマ運動性因子(GMF)の構造解析と腫瘍浸潤における生物学的意義について検討した。2種のグリオーマ由来運動性因子(GMF-I、GMF-II)は、一部相同性を示す癌胎児性フィブロネクチンにおけるED領域の発現において、そのパターンが異なっていた。ED-Bのみを発現していたGMF-Iは、ED-A、ED-B共に発現していたGMF-IIに比べ、T98Gグリオーマ細胞において5-10倍高い運動活性を示した。一方、接着活性については両者に差は見られなかった。T98G細胞以外の種々のグリオーマ細胞や不死化グリア細胞においてもGMF-IはGMF-IIより、高い運動活性を示し、ED領域のスプライシングの違いが運動活性に影響を与えることが示唆された。一方、ラットグリオーマ脳内移植モデルにおいて、グリオーマ細胞とGMFとの共存はグリオーマ細胞単独に比べ、著明なグリオーマ細胞の移動を促進させた。また、抗ED-B抗体とGMF共存下では、腫瘍細胞の移動が抑制された。以上より、GMFがin vitroのみならず、in vivoにおいてもグリオーマ細胞の浸潤に重要な役割を演じていることが示唆された。
