研究概要

バゾプレッシンおよびオキシトシンの生体への生理作用をラットおよびマウス(特に遺伝性多飲マウスを主に)を用いて多方面から解析中である。今年度に得られた成果を以下に列記する。1.ラットにバゾプレッシンのアンチセンスDNAを脳内投与し、飲水量および尿量を観察した。バゾプレッシンのアンチセンスDNAの脳内投与により多飲、多尿が引き起こされた。遺伝性多飲マウスのバゾプレッシン遺伝子の塩基配列を決定した。遺伝性多飲マウスのバゾプレッシン遺伝子の塩基配列は、コントロールマウスとは1塩基のみ異なっていたが、アミノ酸のコードには影響のない部位であった。現在、バゾプレッシン受容体遺伝子の塩基配列を解析中である。2.バゾプレッシンおよびオキシトシンの初期核内遺伝子(hnRNA)をIn situ hybridization法により可視化するために、イントロンを含むcDNAを調整した。遺伝性多飲マウスのバゾプレッシンおよびオキシトシンmRNAの脳内の局在を明らかにした。遺伝性多飲マウスのバゾプレッシンおよびオキシトシンmRNAの脳内の局在は、バゾプレッシンおよびオキシトシンの抗体を用いた免疫組織染色法による細胞体の分布と一致した。バゾプレッシン遺伝子は視床下部で過剰に発現していた。3.パッチクランプ法を用いて、脳スライス標本および単離した神経分泌細胞からの電気記録を行った。カルシウム画像解析装置を用いて、脳スライス標本および単離した神経細胞から細胞内カルシウム濃度の測定法を確立した。パッチクランプ法を用いても、脳スライス標本および単離した神経分泌細胞からバゾプレッシンおよびオキシトシンニューロンに特徴的な電気活動を記録できた。カルシウム画像解析装置を用いて、脳スライス標本および単離した神経細胞のグルタミン酸による細胞内カルシウム濃度の上昇を可視化できた。
