ジーンターゲティングによる疾患モデルマウス作製系の確立

研究概要	マウス胚性幹細胞(ES細胞)と良く似た性質を持ち、且つ培養の容易な胚性腫瘍細胞であるマウスF6細胞と、マウスのトランスサイレチン(ttr)遺伝子に挿入変異を導入する際に作製した置換型ベクターを用いて、微小変異の導入を容易にするベクターの基本構造を確立することが出来た。本年度は、マウスttr遺伝子の5′上流領域から第3イントロンにわたる約6kbのDNA領域を用いて、第2エクソン内にVal30をMetへ置換する変異を導入後、第2イントロン内に、3kb大のイントロン断片の繰り返し配列で挟まれたG418耐性遺伝子(neo)と単純ヘルペスウイルスのチミジンキナーゼ遺伝子(HSV-tk)を選択マーカーとして挿入したベクターを作製した。第一ステップでは、ベクターをF9細胞に導入後、G418耐性細胞を選択し、更にその中からttr遺伝子内に変異を持つクローンをPCR法で選択した。第二ステップでは、得られたクローンから、繰り返し配列内の相同組み換えで、選択マーカーを欠失、ガンシクロビール耐性に変化して、第2エクソン内の変異のみを持つクローンを選択した。この実験で、任意の微小変異を導入可能なベクターの基本構造を確立することが出来た。一方、胚細胞における未分化状態の維持機構の解明に関する研究は難航しているが、この機構に関与する可能性の考えられる多数のcDNAを単離して、構造、機能の解析を進めている。ES細胞とジーンターゲティングの手法を用いて、この中の一つ、我々がrae28と名付けた遺伝子が不活化したマウスの作製に成功した。このマウスは周産期に死亡し、複数の臓器形成に異常をきたすことを見い出しているので、その解析を進めている。
