ジーンターゲティングによる疾患モデルマウス作製系の確立

研究概要	マウス胚性腫瘍細胞株の一つであるF9細胞を用いて分化誘導後、発現が誘導されるcDNAクローンを系統的に単離して、これらのクローンの一つ、Rae28はショウジョウバエで形態形成遺伝子群の発現調節遺伝子として同定されたポリホメオティック遺伝子と相同性を示す事を見い出した。この遺伝子の機能解析をすすめるため、ジーンターゲティングでrae28遺伝子を不活化したマウスを作製、このマウスが多彩な臓器形成の異常を示すことを見いだしたので、本年度は主として、臓器形成の異常を病理解剖学的手法を用いて解析して、以下の結果を得た。ホモ接合体マウスは周産期に死亡し、(i)体重の減少、(ii)目の形成異常、(iii)口蓋裂、(iv)副甲状腺形成不全、(v)胸腺形成不全、(vi)肺動脈狭窄、心室中隔欠損、大動脈狭窄、ファロ-の4徴、などの心臓血管系の異常、(vii)脾臓形成不全、(viii)第7頚椎から肋骨が生じるポステリオール・トランスフォーメーションなどの骨格系の形成異常、などの多彩な臓器の形成異常を示した。これらの症状は、ヒトの先天性遺伝子疾患の一つであるCATCH22症候群の示す複雑な症状と酷似していることが判明した。CATCH22症候群は神経堤細胞の異常によって引き起こされるとされているが、その病因遺伝子や発症機構は不明である。以上の結果、rae28遺伝子のノックアウトマウスはCATCH22症候群の研究にとって絶好の疾患モデルマウスであることが明らかになった。このマウスを利用することで、神経堤細胞と臓器形成との関連を遺伝子レベルで解明可能と考えられる。
