研究概要	申請者らが見いだした細胞間接着装置タイト結合の新しい蛋白7H6抗原(J.Cell Biol.,120:447-483,1993)について、MDCK細胞を用いたATP阻害実験で示唆された7H6抗原の局在と、タイト結合のバリアー機能の相関(Exp.Cell Res.,222:269-274,1996)を、ラット肺血管内皮(RLE)細胞株、培養ニワトリ網膜色素上皮細胞、ニワトリ消化管粘膜上皮の攝餌に伴う発育過程、初代培養ラット腹膜中皮細胞を用いて研究した。その結果、これらの異なった動物種、細胞種の、異なった実験条件の系においても、7H6抗原のタイト結合への局在とバリアー機能の増強が良く相関していることが確かめられた。血管内皮や網膜中皮細胞におけるバリアー機能の増強は、がん細胞の血管外脱出や中皮細胞下への侵入を有意に抑制することから、タイト結合のバリアー機能を増強する7H6抗原の発現調節の研究は、がんの転移、腹膜播種の抑制にも役立つ可能性が期待される。また消化管におけるバリヤ-機能の低下は、細胞の侵入を容易にし、敗血症、エンドトキシン血症を招くことから、その調節に関与する7H6抗原の解析が重要な研究課題となる。このような観点から7H6cDNAのクローニングを進め、その塩基配列から分子構造の解析を行った。その結果7H6抗原は、マイコプラスマからアフリカツメガエルまでに広く存在し、染色体の凝集に働くSMCファミリーの蛋白と相同であることが明らかにされた。SMC蛋白はATP結合部位を持ち、分子モーターとして働くことが推定されており、7H6抗原がATP存在下にモーターとして働き、タイト結合の構成蛋白のコンフォメーションあるいは配列を変えて、バリアー機能を調節している可能性が示唆された。細胞間コミュニケーションに働くギャップ結合と心筋細胞の周期性拍動の関係についても新しい知見を得た。
