研究概要	変異体アッセイsystemの改編;大腸菌supF遺伝子は、終止ゴドンUAGをチロシンと読み替えるアンバーサプレッサーtRNAを作る遺伝子である。supF^+AEsupF^-の変異を鋭敏に検出するために、大腸菌宿主を以下の手順で改良改変した。抗生物質ナリジクシン酸(nal)抵抗性gyrA遺伝子のアンバー変異、乳糖分解lacZ遺伝子のアンバー変異、抗生物質ストレプトマイシン(Sm)抵抗遺伝子rpsLのアンバー変異の3種類のアンバー変異を持つプラスミドを作成し、三重の選択をできるようにし、変異体同定を一層簡便化した。この系での自然突然変異頻度は5x10^<-8>〜5x10^<-7>であった。子の系を用いて、鉄イオンの細胞への取り込み変異が、8-ヒドロキシグアニン由来の変異を多数誘発することを明らかにした。哺乳類培養細胞やマウスへのsupF遺伝子の導入を行っている。実験マニュアルを作成した。活性酸素誘発突然変異のスペクトル;上記実験系を用い、チミングリコール等の活性酸素性塩基損傷修復に関わる大腸菌遺伝子、nthおよびneiの変異株での自然突然変異を調べた。GCAEATの変異が大部分を占め、5-ヒドロキシシトシンも重要な変異塩基損傷である可能性を指摘した。コバルト(II)はDNAに直接結合する可能性が指摘されていたが、DNA-Co(II)複合体形成に依存し、欠失突然変異が増大していた。
