抗DNA抗体増強因子ヌクレオバインディンと自己免疫-その病態と分子細胞機構の解明

研究概要

4リットルスケールの大腸菌培養で約5mgのヌクレオバインデイン(Nuc)を回収出来る精製法が確立された。抗体カラムによるone-step精製法の樹立は現在進行中である。
Nuc(50μg)の正常マウス腹腔内投与後15時間で再現性をもって胸腺細胞のアポトーシスと血中ヌクレオソームの出現が観察されるが、抗CD3抗体を同時投与すると、何れの単独投与を遥に凌駕した胸腺細胞のアポトーシスと血中ヌクレオソームの出現がみられた。血中に出現するヌクレオソームDNAは同時投与の場合その停滞時間の延長がみられ、NucがヌクレオソームDNAと結合し安定化することが示唆された。SLEを自然発症しているMRL/lprマウスからも同様のヌクレオソームDNA同定され、Nucの血中濃度の上昇が自己免疫の発症または維持に重要であることが分かった。Nuc投与直後に於ける胸腺細胞蛋白質分子の動態をSDS-PAGE及びイムノブロットにて解析し、ヒストンH1分子の増加、38-40kDa蛋白質のチョロシン燐酸化、50kDa蛋白質の新たなチョロシン燐酸化が観察された。これらの所見はNucが胸腺細胞と交信したことを示すもので、その本態の解明が進んでいる。
正常マウスへのNucの間歇投与で誘導される高ガンマグロブリン血症はMRL/lprマウス脾細胞培養にNucを添加しても再現され、培養上清中にヌクレオソームDNAの増加が認められたことから、Nucが細胞外に放出されたヌクレオソームDNAを安定化し、ポロクローナルB細胞活性化を誘起することが示唆された。
