Gタンパク質共役受容体のリン酸化と脱感受性

研究概要

CHO細胞(Chinese Hamster Ovary Cells)にムスカリン受容体m2サブタイプ(m2受容体)、細胞内第三ループを欠損させたm2受容体(I3-delm2受容体)、Gタンパク質共役受容体キナーゼ2(GRK2)を安定的に発現させ、m2受容体の脱感受性(Desensitization)、細胞内移行(Internalization)、消失(Down Regulation)を調べた。内向き整流KチャネルGRK1とCRK1を一過性に発現させたCHO細胞で、心房細胞と同様のカルバミルコリン依存性K電流が測定できた。GRK2を共発現しない細胞では脱感受性が見られなかったが、GRK2を共発現した細胞では数分以内の早い経過で起こる脱感受性現象が見られた。I3-del m2受容体とGRK2を発現した細胞では脱感受性は観察されなかった。CHO細胞を種々の濃度のカルバミルコリンで刺激すると、濃度依存的な受容体の細胞内移行と消失が観測された。GRK2を共発現した細胞では、低濃度のカルバミルコリンでの細胞内移行と消失が促進され、濃度依存曲線が左側にシフトした。カルバミルコリンで刺激し続けると、80-90%の受容体がt_<1/2>=9.5minで細胞内移行し、60-70%がt_<1/2>=2.3hで消失した。I3-del m2受容体では細胞内移行がt_<1/2>=8.4h、消失がt_<1/2>=9.9hと大幅に遅くなった。これらの結果は、GRK2によるリン酸化が受容体の脱感受性と早い細胞内移行に必要で、細胞内移行に接続して消失が起こることを示唆している。
