Single-chain抗体とnaked DNAによる難治性C型肝炎の治療

研究概要

B型肝炎ウイルス(HBV)のコア蛋白(HBc)に対する単鎖抗体(sFv)1C9を、またHBCと反応しないコントロールの単鎖抗体γ9も作製した。これら単鎖抗体の効果をHBV粒子産生肝癌細胞株HB611で検討した。すなわち、1C9およびγ9発現HB611細胞株を樹立し、HBV転写産物・蛋白の産生、HBV複製中間体およびHBV粒子の産生に対する影響を検討した。その結果、HBV転写産物・HBc発現量に差を認めなかったが、HBV複製中間体はγ9産生株に比し1C9産生株で抑制されていた。このことより、HBcに対する単鎖抗体は、HBV-RNAのHBc内へのパッケージングを抑制することによってHBV複製を制御すると考えられた。以上のことはHCVの場合にも応用可能であり、種々のHCVコア蛋白に対するモノクローナル抗体を作製し、単鎖抗体をクローニングすることより、HCV-RNAをコア蛋白へのパッケージングの段階で抑制可能と考えられた。これまで多くの施設で、アンチセンスオリゴやリボザイムによる転写レベルでの抑制実験は報告されていたが、今回我々はウイルス粒子の形成段階で抑制が可能であることを示した。
