肝病態迅速把握のための血中ケトン体(比)高感度測定法の開発

研究概要

平成7年度、NADHオキシダーゼを用いる高感度で迅速に血中ケトン体を分別定量法、すなわち、β-ヒドロキシ酪酸脱水素酵素(β-HBDH)反応でβ-ヒドロキシ酪酸(β-HBA)濃度に比例して増加するNADHにNADHオキシダーゼを作用させて過酸化水素を生成させ、同時にPeroxidase共存下で、3-metyl-2-benzothiazolinone-hydrazone-6-sulfate(MBTH-S)とN-Ethyl-N-sulfopropyl-3,5-dimethylanilineを酸化縮合発色させ、波長570nmに極大吸収をもつ高感度な発色生成物の発色増加量よりβ-HBAを求め、またアセト酢酸はβ-HBDH酵素反応でβ-HBAに変換し、血中の既存のβ-HBAとの総和として測定した後、既存のβ-HBA分を差し引いて求める方法を確立したが、そのより高感度化の展開として、MBTH誘導体(カップラー)とTrinder試薬(水素供与体)の組み合わせ、反応条件について、1)現組み合わせでの反応条件変更での感度への影響と、2)新規MBTH誘導体として、理論上1分子過酸化水素で発色が期待できるヒドラゾノ基に脱離基導入の2-[(methylsulfonyl)hydrazone]-2,3-dihydro-3-methyl-6-benzothiazole-sulfonic acid、及び水溶液での安定性向上が期待される2-hydrazone-2,3-dihydro-3-methyl-6-benzo-thiazolesulfonyl)aminoacetic acidをそれぞれ合成し、感度に及ぼす影響を検討した。その結果、1)ではトリス緩衝液を用い、pHを8付近とすることにより、現条件での約2倍の感度が得られた。しかしながら、試薬ブランクの上昇度が現条件に比し大きく、精度面への悪影響があるため、この点について現状、改善検討中にある。2)についてはいずれの化合物も期待する感度、安定性を得ることができなかった。今後は1)の改善(精度の確保)後、次のステップに研究を進める予定である。
