IL-12によるT細胞亜集団Th1の分化機構の解明

研究概要

細胞性免疫を制御するT細胞亜集団Th1の分化・活性化は主としてIL-12によって制御されている。従って、本研究ではIL-12の産生およびその制御、IL-12受容体発現の制御に的を絞って立案した。IL-12はP35とP40から成るヘテロダイマーである。抗原提示細胞によるp40の産生はCD40刺激によって起こる事は、我々を含む複数の研究グループによって既に明らかにされている。本研究ではp35の産生を誘導する抗原提示細胞上の分子の特定を試みた。その結果、MHCクラスII分子が該当する事が明らかになった。MHCクラスII分子刺激によって産生されるp35はCD40刺激によって産生されるp40と共に生物学的活性をもつIL-12p70を構成し、細胞外に分泌されることもわかった。IL-12受容体もβ1、β2のヘトロダイマーである。これは共にTCRおよびB7刺激によって発現されるが、とくに、β2の発現にはB7刺激が重要である。ナイーブTh細胞ではβ2の発現にはB7を介する刺激が必須である。B7刺激を受容するTh細胞上の分子としてはCD28とCD152があるが、前者はIL-12受容体、特にβ2の発現には促進的に働き、後者は抑制的に作用する事も明らかになった。ナイーブT細胞からTh1が分化する機構にはIL-12非依存性経路がある事も判明した。この経路はCD152によって誘導される。この経路は3つからなる。TGF-βの産生を刺激してTh1分化を促進し、IL-4産生増強によってTh2分化を抑制する。さらに、液性因子を介さない未知の経路も存在する事も明らかになった。この経路の解明は今後の課題である。IL-12によるTh1細胞分化・活性化が生体内でもみられるか否かも検討した。このために、IL-12p40トランスジェニックマウスも作成し、マウスマラリアモデルを用いて検討し、IL-12はTh1を誘導して、感染防御に免疫に寄与していることを確認した。
