研究概要

1. 安全なマテリアルを作成するための準備
Naked DNA法のマテリアルとしてplasmid DNAを作成し、エンドトキシン等を定量し安全性を確認した。
DNAのdegradationを防ぐために糖鎖にてDNAを修飾することにより、DNAase耐性ののマテリアルを作ることに成功した。
昨年秋に完成した名古屋大学医学部・遺伝子調製室を用いてマテリアル管理をGLPレベルにて行うことを準備中である。
2. 非ウイルスベクターによる効率の良い遺伝子導入法の開発
SFVの構造を基本構造とした新しいDNA型発現 vector、pSFV3-CMV-pAを作製した。このvectorはSFV由来のreplicaseの機能を介し、哺乳動物細胞内で外来遺伝子産物を大量に発現した。従来のSFV由来の発現vectorと異なり、in vitro transcriptionによるrecombinant RNAの合成が不要で、p1asmid DNAのままで遺伝子導入に用いることが可能である。導入後は一過性に大量のタンパクを産生し、その後導入細胞は死滅する。この特徴を生かした遺伝子免疫法などへの応用が検討している。
3. 実際の臨床に向けて、患者登録、プロトコールの作成
既に患者検体を100例近く集め、遺伝子治療の適応になる患者の遺伝子配列、血清、細胞保存を開始している。臨床プロトコールの作成も進んでおり現在のVersionを研究成果報告書に添付している。
