分泌シグナルを持たない増殖因子の細胞外輸送機構の解析

研究概要

明確な分泌シグナルを持たないためその細胞外移行のメカニズムが明らかにされていない塩基性線維芽細胞増殖因子(bFGF)とその可溶性受容体およびクリプト(CR-1)を発現する細胞株を樹立しこれらの分子がどのような分布を示して細胞外へと輸送され、また細胞表面から遊離されるのか、その機構と経路を解析した。
bFGFについてはその可溶性受容体の共発現系をマウス正常線維芽細胞BALB/c3T3を用いて確立し、この形質転換細胞が培地中にbFGFを分泌することを確認した。これはbFGFを単独で発現させても観察できない現象であり、bFGFの細胞外移行に可溶性受容体が関与している可能性を示した。また、上皮細胞成長因子のモチーフをもつ新しい増殖因子CR-1について、種々の方法でその性質を解析したが、細胞で発現したCR-1はウエスタンブロッティングで20、22、24、28Kdの分子量を示した。遺伝子の配列から分泌シグナルを予測すると36アミノ酸がプロセスされる結果、約17Kdの分子量になると考えられたが、上の結果からはシグナルのプロセッシングが行われていないことを示唆するものである。抗体を利用して定量を試みると培養上清中には100ng/mLのCR-1が分泌されていることを確認しCR-1が通常のレベルで分泌されていることがわかった。さらに、グリーンフルオレッセントプロティン(GFP)との融合タンパク質をデザインし、細胞中の蛍光の局所的変化をを解析したが、明確な差を解析するに至っていない。しかし、さらに研究を進める上で重要な知見を積み重ねることができた。近年CR-1は発生段階で非常に重要な役割を果たしていることが示唆され、本研究の重要性はさらに高まっている。
