研究概要

須田のクローニングしたマウス幹細胞抗原CD34分子に対する抗体を用いて、中内は、自己複製能をもつ幹細胞ともたない前駆細胞を区別できることを、また、骨髄有核細胞中0.004%の頻度でしか存在しないCD34陰性造血幹細胞を純化し、その細胞1個でマウス個体の造血を再建し長期間維持できることを明かにした。これは「真の造血幹細胞」を単離できた画期的な研究成果であり、この幹細胞の未分化維持機構などの分子基盤を明らかにすべく、世界中の研究者が集中しつつある。西川・須田は、種々の受容体型チロシンキナーゼに対する抗体を作成して、血液細胞と血管内皮細胞の共通祖先細胞であるHemangioblastを同定した。AGM(Aorta:大動脈,Gonad:生殖器,Mesonephros中腎)領域が二次造血の始まりであることを示し、造血システムの発生に関する研究は、その誘導を可能にする微小環境・分子基盤の解析を含めて大きな発展がみられた。ことに、造血幹細胞と血管内皮細胞の発生における転写制御とそれらの機能的な相互作用は、造血、血管新生の機構を明らかにすると考えられる。中畑は、幹細胞の効率的な対外増幅に成功している。可溶性IL-6受容体を細胞外から補充することによりgp130からのシグナルを強制的に造血幹細胞増幅に利用するという方法は、きわめて論理的な増幅法であり、臨床応用への展開も期待されている。
