研究概要

小児固形腫瘍は分化・成熟過程の胎児性組織を発生母地とし、個体や器官形成を模倣する。このような腫瘍群の生物学的特性や発症機構を解析し、器官形成機序と腫瘍化との関連を以下のように明らかにした。
1.ヒト胎児性癌細胞の初期分化過程で新たに単離されたEATの個体での機能を知るため、トランスジェニックマウスの作出に成功した。遺伝子導入マウスを用いて種々解析した結果、EATがシスプラチン投与に起因するアポトーシスによる腎尿細管壊死を阻止した。また、マウスEATをクローニングし、マウス初期胚でのEATの消長を解析した結果、EATは受精直後の4-8細胞期で最も発現が亢進することが明らかになった。以上の結果より、EATは初期発生の維持に重要な分子であることが明確になった。
2.胎児性癌特異抗体4C4の認識する抗原を明らかにした。
3.10余例の散発性肝芽腫を用いて、その発癌機序を11p11.5に集積するインプリンティング遺伝子の異常という見地から解析した。その結果、インプリンティング遺伝子H19のLOHまたはloss of expressionによる不活化が肝芽腫発生に最も重要であることが明らかとなった。
4.散発性および形成異常を伴ったウイルムス腫瘍のWT1遺伝子変異を解析した結果、DrashおよびFrasier症候群に特徴的な変異を同定した。
