研究概要

以下研究課題ごとに成果を記す。
1) 温度感受性葉緑素突然変異体:
イネの温度感受性葉緑素突然変異体の形質発現を明らかにする目的で、ファイトトロンを利用し、温度の効果を再確認した。すなわち最低気温に反応して白色部が発現すること、処理期間が3-4日間で発現を始め、処理が長くなるに従って、白色部が大きくなることが確かめられた。また、処理温度が19から14℃で徐々にはっきりした白色になることが確かめられた。
2) 根の発生分化と形態形成に関わる突然変異体:
これまでに得られている根の発生分化と形態形成に関わる多数の突然変異体を用いて遺伝解析を行い、遺伝子座の同定とアレリズム及び変異体遺伝子間の相互作用性の有無を明らかにした。これらのうち数種の変異体に着目して変異体xカサラスのF2集団を作成し、原因遺伝子のファインマップ作成のため分子マーカーによる連鎖解析を進めた。また、根の物理刺激応答性変異体BRX180を同定し、ホルモン反応の側面から特性解析を行った。
3) 恒常的ジベレリン感受型長稈突然変異体:
この変異体がジベレリンのシグナル伝達の異常であることを実験的に証明した。また、原因遺伝子のクローニングにほぼ成功した。今後この遺伝子の構造・機能の解析を進める予定である。
4) FISHによる効率的マッピング法の開発:
各変異体の原因遺伝子の物理地図作成に用いる、RFLPマーカー、CAPSマーカーおよびRAPDマーカーをプローブとした蛍光in situ hybridization法の効率化について、技術上の検討を行っている。特に、原因遺伝子の密接連鎖マーカーの効率的オーダリングのために、検出するクローンのサイズおよびコピー数の調整を行っているところである。
