水分恒常性の維持機構の分子細胞生物学的研究(神経内分泌細胞間・内情報伝達の解析)

研究概要

生体の恒常性を保持する中枢である視床下部において室傍核および視索上核が体液の恒常性を維持するのに中心的役割を担っていることが明らかになってきた。また分子生物学的手法および電気生理学的手法により神経性・内分泌性および局所での細胞間情報伝達、またそれによる細胞の反応などが徐々に解明されてきている。昨年度、PACAP、プロスタグランデインE2、アドレノメデユリンなどの神経分泌細胞への作用について明らかにしてきた。また、この室傍核および視索上核には抗利尿作用を有するバゾプレッシン産生細胞があり、これらの神経分泌細胞は他の神経ペプチドや神経型NO合成酵素を含有している。昨年度は、糖尿病などの高浸透圧状態や循環容量の減少により、室傍核および視索上核の神経分泌細胞で神経型NO合成酵素遺伝子が増加すること報告した。これらの結果を踏まえ、今年度得られた業績は以下の通りである。1、プロスタグランデインE2はシナプス前のEP受容体を介して、神経分泌細胞へ興奮性に働く系があることを確認した。2、昨年報告した、ラット視索上核の神経分泌細胞のGABAB受容体を介した神経活動の抑制は、カルシウム電流の抑制によるものであることが解明された。3、アデノシンはラット視索上核の神経分泌細胞のA1受容体を介してカルシウム電流を抑制した。4、我々が継代培養している遺伝性多飲マウスの視床下部においてバゾプレッシン遺伝子が増加していることを解明した。5、麻酔薬として臨床応用されているプロポフォールが視索上核の神経分泌細胞を抑制することを解明した。6、室傍核および視索上核での神経型NO合成酵素遺伝子の絶食による減少がレプチンの脳室内投与による補充により回復することを明らかにした。7、NOが視索上核の神経分泌細胞においてクロライド電流を変化させることを解明した。
