研究概要

生物活性化合物の構造、特に絶対配置を含めた立体化学は、基本的な情報として重要である。本研究では分子キラリティーの情報をデータベース化し、インターネットに乗せて全世界に発信する研究を行っている。分子キラリティーのデータとして、1)旋光性および円二色性と絶対立体化学のデータ、2)X線結晶解析と絶対構造のデータ、3)NMRによるキラル認識と絶対立体化学のデータ、4)キラルHPLCによる光学分割のデータ、5)キラルプールやキラル補助基のデータ等をデータベース化する。また、データベースを各国の研究者が容易に閲覧できるためのインターネット化の環境整備を行う。
1.我々はキラル化合物のCDとUVスペクトルおよび絶対立体化学のデータベース化を世界で初めて行ったが、それをさらに発展させた。CDスペクトルの帰属と解析にはUVスペクトルの情報は必須であり、両スペクトルを図形として同時に見ることを可能にした。また化合物の絶対立体化学は図形として表示し、容易に認識出来るようにした。また、化合物とスぺクトルのキーワード、スペクトルの測定条件、文献などを入れられるようにした。
2.データベ-スを公開するには、インターネットが最適である。このため、データ集積用高速ワークステーションの2台目を導入し、スピードアップをさらにはかった。
3.絶対配置を明確に決定する方法としてX線結晶解析が最も有用である。我々はキラル化合物のX線結晶解析データの登録を開始した。絶対配置決定の基本データとして重要である。
4.我々は本データベースの作成に際して信頼あるCDとUVスペクトルデータを登録することにしている。このため、世界中のCD研究者に呼びかけ、データの提供を受ける環境整備を行った。また、サンプルの提供を受け当研究室で測定して登録を行っている。
