ヒト細胞導入モデルとしてのリンパ球完全欠損マウスの利用

研究概要

ヒトにおける疾患の研究で、自然発生や人工改変の変異マウスが動物モデルとして多く用いられている。我々はインタロイキン2(IL-2)の受容体の第3の構成成分であるγ鎖のクローニングと、ジーンターゲッティング法によりγ鎖欠損マウスの樹立に成功した。γ鎖遺伝子欠損マウスとRAG2遺伝子欠損マウスをかけ合わせることによって、リンパ球(T細胞・B細胞・NK細胞)を完全に欠失するマウスを作出し、その性質を調べた。リンパ球完全欠損マウスをヒト細胞の導入宿主として確立する実験を行った。
γ鎖欠損マウスを遺伝的背景をC57BL/6とBalb/cにするためバッククロスを行い、すでに8代のバッククロスが完了している。またγ鎖遺伝子、RAG-2遺伝子の両方を欠損しているマウスのバックグランドも完全にC57BL/6とBalb/c遺伝的背景になっている。今回、C57BL/6バックグランドのリンパ球完全欠損マウスにxenograftとなるヒトの大腸癌の手術摘出サンプルを移植して、経過を観察した。大腸癌切除標本のうち、肉眼的にviabilityが高い部分の約5mm角をマウス皮下に挿入移植し、肉眼的・顕微鏡的に検討した。
現在までに、7例のヒト大腸癌をマウスに移植し、死亡した1例を除いた6例で、移植後2〜6か月を経てマウス皮下に腫瘤が残存しており、腫瘤は生着している。顕微鏡観察で1例は、腫瘍細胞が筋層内・血管内へ浸潤していた。
以上の結果は、ヒト大腸癌がマウスに生着し、浸潤を起こし得ることが示し、リンパ球完全欠損マウスがヒト細胞の受容体として機能することが明らかとなった。
