血液・脳関門の有機アニオン透過の分子機序の解明-モノアミン代謝物及び薬物抱合体排出のメカニズム

研究概要

血液・脳関門の有機アニオン輸送系の分子的実体を明らかにする目的で、ラット腎由来の多選択性有機アニオントランスポーターOAT1及び多選択性中性アミノ酸トランスポーターLAT1の塩基配列を基にしたホモロジークローニングを行った。EST(expressed sequence tag)データーベースから得られた塩基配列を基に、有機アニオントランスポーターOAT1と相同姓のある新規トランスポーターOAT4を同定した。OAT4は、OAT1、OAT2、OAT3と38〜44%の相同性を有し、腎及び胎盤に発現が見られた。アフリカツメガエル卵母細胞に発現されることにより、OAT4はestron sulfate(Km:1.01 μM)及びdehydroepiandrosterone sulfate(Km:0.63 μM)を高親和性に輸送することが明らかになった。OAT4は、さらに種々の化合物の硫酸抱合体を受け入れ、硫酸抱合を受けた内因性物質あるいは薬物や外来性異物の細胞膜透過に重要な役割を果たすと考えられる。多選択性中性アミノ酸トランスポーターLAT1の塩基配列を基にしてホモロジークローニングにより、各種新規トランスポーターすなわち中性アミノ酸トランスポーターLAT2、中性及び塩基性アミノ酸を基質とするBAT1、小型の中性アミノ酸を輸送するAsc-1を同定した。このうちLAT2とAsc-1が脳に強く発現していた。特異抗体の得られた、LAT1、LAT2、BAT1、Asc-1及びそれらの機能発現に必須な補助因子4F2hcについて、血液・脳関門の構成要素である脳毛細血管内皮細胞における発現を検討したところLAT1、LAT2、及び4F2hcが脳毛細血管内皮細胞に存在することが明らかになった。各種有機アニオントランスポーターOAT1、OAT2、OAT3、及びOAT4の特異抗体を用いた脳組織の免疫組織化学的検討が現在進行中である。
