ノックアウトマウスを用いたIL-1の免疫反応及びストレス応答における機能解析

研究概要	本年度は我々の作製したIL-1関連遺伝子群KOマウスを用いて、炎症反応と抗体産生におけるIL-1の役割について解析した。LPSによるエンドトキシンショックに対する感受性を調べたところ、致死量のLPSに対してIL-1KOマウスは野生型と差がないのに対して、IL-1raKOマウスが高感受性を示し、II-1raは防御的な役割を果たしていることがわかった。またP.acnesで感作して低力価のLPSで劇症肝炎を引き起こす系ではIL-1KO及びIL-1raKOマウス共に高感受性を示した。更にzymosanによる皮膚炎やConA誘導肝炎においてもIL-1KOマウスではその発症が抑制されたことから、これらの炎症反応にもIL-1が重要な役割を果たしていることがわかった。次に通常飼育条件下での血中の免疫グロブリン濃度を調べたところIL-1KOマウスでは低下し、IL-1受容体アンタゴニスト(ra)KOマウスでは亢進していることが見られ、胸腺依存性抗原であるSRBCで免疫した場合も、IL-1KOマウスではIgMとIgGの抗体産生が抑制され、IL-1raKOマウスでは亢進していることがわかった。このことからIL-1とIL-1raが抗体産生を調整していることが明らかとなった。マイトジェンや抗IgM刺激に対するTやB細胞の反応性には異常がなかったが、抗原特異的なT細胞増殖応答には差が認められたので、T細胞とB細胞や抗原提示細胞との相互作用にIL-1が重要な役割を果たしていることが示唆された。IL-1raKOマウスをBALB/c背景にすると、高率に関節炎を自然発症することがわかった。関節の病理像はヒトの関節リウマチとよくにており、リウマチ因子や抗コラーゲン抗体、抗DNA抗体の産生も認められ、新しいリウマチのモデル動物を作製することができた。
