研究概要	SV40やAAVのカプシドタンパク質はVP1、VP2、VP3の3種類があり、野性型ウイルス粒子のの主要なコンポーネントは、SA40ではVP1、AAVではVP3である。我々は、これまでにバキュロウイルス発現系を用いて、SV40のカプシドタンパク質VP1を単独発現した場合と、AAVのカプシドタンパク質の場合はVP2とVP3の両方を共発現したときに昆虫細胞内にウイルス粒子様粒子(VLP)が形成されることを明らかにしてきた。またVLPを回収率よく精製し、それを試験管内で分散させて粒子を再構築させるのに必要な諸因子を明らかにしてきた。それらカプシドタンパク質を遺伝子工学的に改変を行って、それらの再分散化や集合化に関する機能ドメインをアミノ酸レベルで解明した。また細胞染色体への組み込みなどベクターとして要求される機能を持つ組み換えプラスミドが、カプシドタンパク質から成るカプセル内に効率よく包含される諸条件を検討した。(J.Virology,1999.p7912-7915)
