研究概要	SV40やAAVのウイルス粒子は、それぞれ主要なカプシドタンパク質であるVP1およびVP3によって外側を被覆されている。これらをバキュロウイルス発現系を用いて昆虫細胞内で発現すると、SV40ではVP1のみでウイルス様粒子(VLP)が形成され、AAVでは核移行シグナルを付加するとVP3のみでVLPが形成されることを見出した。そのSV40のVLPを回収効率良く精製し、試験管内でペンタマーにまで解離、分散し、VLPを効率良く再構成する条件を設定した。その再構成には、カルシウムイオンとその結合部位およびヂスルフィド結合が関与することを見出し、それに関与するいくつかのドメインおよびアミノ酸残基を決定した(J.Virol.,75,61-72,2001)。また、その中にプラスミドDNAを包含するために、DNAコンパクションに関与すると示唆されているVP2やVP3の組換えタンパク質を大量に調製し、それらの機能を検討する実験系を確立した。さらに、新規機能を付加したVLPを構築するために、VP1の改変体を遺伝子工学的に作製して、それらのVLP形成能を検討した結果、VP1は極めて堅い構造をしたタンパク質であるが、粒子表面に露出されるいくつかの部位の中から新規機能を付加することが可能である部位を見出した。
